I know obviously this story takes place before Dumbledore finally defeats Grindelwald, but is it supposed to be taking place before or after the fight that killed his sister, Ariana?

I'm pretty sure it's after Ariana's death, because at the time of her death Grindelwald wasn't known yet. He and Dumbledore were discussing their theories, but Grindelwald hadn't actually done anything yet.

It takes place about 28 years after her death.
